Solution

pBR322 has two antibiotic-resistant genes for ampicillin (ampR) and tetracycline (tetR). Due to the insertion of the gene in the ampR region, insertional inactivation results in the inactivation of the ampicillin gene and hence the organisms become sensitive to this antibiotic and do not grow in the media containing this antibiotic.
