Solution

T is an unmarried male and U (P's daughter) is female; T and U are the two unmarried siblings, children of a couple. The parent couple are P and Q. Q is R's mother-in-law, so R married a child of Q. Since T and U are unmarried siblings, R's spouse must be S. S is male, so R is female and R is S's wife.
